    多くの解剖学的・生理学的・分子生物学的研究によって、小脳は正中線を軸にして内側から外側方向へ左右対称の縦縞状の区画に領域化されていることが示された。そこで、小脳の縦縞状の領域は小脳における機能区分と考えられ、小脳の神経回路網の形成と小脳の機能発現における基礎的構造単位であると考えられるようになった。しかし、最も基本的な疑問である、「小脳の領域化が、いつ起こり、何によって規定されているか。」は、解明されていなかった。そこで、アデノウイルスベクターによる神経細胞の誕生日特異的遺伝子導入法を用い、小脳における縦縞状領域の形成過程を解析した。その結果、小脳の縦縞状領域化は、小脳プルキンエ細胞の発生する時期(誕生日)に依存して形成されることが判明した。アデノウイルスベクターを用いた遺伝子導入は、神経細胞の発生・分化、脳の機能を解析する上で大変有利である。この技術を用いれば、小脳において、ある特定の縦縞状領域に遺伝子を導入することができる。我々は、薬剤によって神経細胞を殺すことのできる遺伝子を発現するアデノウイルスベクターならびに、神経細胞の生理的活性を変化させる遺伝子を発現するアデノウイルスベクターを作製した。前者のアデノウイルスベクターを用いれば、同一の誕生日を有するプルキンエ細胞群を殺すことができる。または、後者のアデノウイルスベクターを用いれば、ある特定の縦縞状領域の生理活性を人...

    多くの哺乳動物においてフェロモンを介した動物行動学的現象は数多く報告されているが、フェロモンがどのような分子でどのように認識されているかなど、分子生物学的機構は不明な点が多い。近年、分子生物学の発展により、ラットならびにマウスの鋤鼻器よりフェロモン受容体の候補がクローニングされた。現在、構造の異なる2つのグループのフェロモン受容体が報告されており、1つのグループは、匂いの受容体に構造が類似しており、もう1つのグループは、代謝型グルタミン酸受容体に構造が類似している。後者のグループに属するVR1ならびにVR4を特異的に認識する抗体の作製を試みた。VR1ならびにVR4のC-末端と同一のペプチドを合成し、それらを抗原としてウサギに免役することによりポリクローナル抗体を作製した。作製した抗体を用いて、マウス成体の鋤鼻器を免疫染色したところ、それぞれの抗体は鋤鼻器の神経細胞を特異的に染色し、また、その神経細胞は鋤鼻器の基底部に局在していることが明らかとなった。一方、これらの抗体によるマウス鋤鼻器における陽性反応は、マウス胎仔では観察できず、出生後、哺乳されたマウスの仔では陽性反応が観察できた。出生後すぐのマウスは、目が見えず耳も閉じており、外界からの情報収集は主に匂いまたはフェロモンに依存している。特に出生後すぐの仔は、匂いまたはフェロモンによって母親を認識し、母親の乳首を探索し、哺乳...
