    申請者らは、食道扁平上皮癌において術前化学療法により腫瘍随伴マクロファージの浸潤が促進すること、また腫瘍随伴マクロファージの浸潤にはIL-34が関与する可能性を見出しており、術前化学療法により食道扁平上皮癌細胞から産生されるIL-34が治療効果や予後に負の影響を及ぼす可能性が推測される。そこで本研究では、以下3点を中心に検討を行う。
    ①食道扁平上皮癌組織のIL-34産生レベル又は腫瘍随伴マクロファージの浸潤レベルが術前化学療法の治療効果および予後に及ぼす影響の検討
    ② 食道扁平上皮癌細胞の抗癌剤感受性とIL-34産生レベルの相関に関する検討
    ③ 抗癌剤で処理した食道扁平上皮癌細胞がマクロファージの分化、機能に及ぼす影響の解析
    本年度は、食道扁平上皮癌細胞のIL-34産生レベルが化学療法の治療効果や予後に及ぼす影響に関し、食道扁平上皮癌組織切片を用いた解析を実施した。具体的には、術前化学療法施行後の食道扁平上皮癌組織においてIL-34産生レベルを中央値によりIL-34高値群とIL-34低値群に分類し、それぞれの治療効果判定や治療後の全生存期間、再発に関する検討を行った。IL-34低値群と比較し、IL-34高値群において治療効果判定が不良(Grade 0-Ia)な患者の割合が多く見られ、生存期間が短く、再発の割合が高いという結果が得られた。したがって食道扁平上皮癌において術前化学療法が誘導するIL-34が治療効果を低下させ、予後を悪化させる可能性が示唆された。

    In this study, we revealed that radiation therapy induced M2-polarization of tumor-associated macrophages (TAMs) in esophageal squamous cell carcinoma (ESCC). The number of CD163+ M2-TAMs was increased by radiation therapy in ESCC tissues. Radiation also increased the expression of CD163, PD-L1 and PD-L2 in human peripheral blood mononuclear cell-derived M2-macrophages (called TAM-like cells). Moreover, we found that radiation triggered the expression of IL-34 in ESCC cells, which further enhanced M2-polarization of TAMs.

    In this study, we investigate whether and how obesity exacerbates allergic reaction.
    We found that increased plasma histamine level by passive systemic anaphylaxis, a classical model of IgE/mast cell-mediated systemic allergic reaction, was significantly higher in high fat diet-induced obese mice than non-obese mice. To elucidate how obesity reinforces allergic reaction, we focus on the roles of adipose tissue macrophages because they are closely associated with obesity-related chronic diseases. In vitro experiment revealed that mRNA expression of osteopontin in macrophages was up-regulated by co-culturing with adipocytes. It has been reported that osteopontin regulates IL-12 production. Indeed, we found that IL-12 significantly enhanced degranulation of mast cell by antigen-challenge.
